If you insulation extends above the joists (which is often the case if the full currently recommended 270mm depth of insulation is fitted , especially in older properties), flooring placed directly over insulation risks compressing the insulation, reducing its efficiency and issues caused by inadequate ventilation. In such cases, raising the loft floor level with a wooden subframe will allow you to take advantage of both full benefits of insulation and the safer, easier access to an extended storage area created by boarding the loft.

Of course. You decide how much of your loft you want boarded, and this will depend on what you want to use to loft space for, how big it is in the first place, the roof height, and other factors. If all you need is a place for a few boxes, or easier access to installations (e.g. water tank or piping) in the loft space, partial boarding might be enough. On the other hand, if you prefer to create a luxury storage room with cupboards, windows, lighting and comfortable access -- in other words a non-habitable loft conversion -- you will likely get the whole floor area boarded. When making this decision, remember that the amount of stuff to be store is likely to increase every year, and that you will need free floor space for walking through your storage space.
